Human Epididymis Protein 4 (HE4) belongs to the whey acid 4-disulfide center (WFDC) protein family and has the properties of a trypsin inhibitor. The HE4 gene is located on chromosome 20q12-13.1 of the human genome and encodes a 13 kDa protein that reaches 20 kDa to 25 kDa during its mature glycosylation and includes a single chain containing two WFDC domains. Human epididymal protein 4 is a new ovarian tumor marker, which can help clinically detect ovarian cancer at an early stage, and has certain guiding significance for its risk assessment. This assay can be used as an adjunct to monitoring disease recurrence and progression in patients with epithelial ovarian cancer. HE4 has the highest sensitivity for ovarian cancer detection, especially as a disease in the early asymptomatic stage, when CA125 and HE4 are combined, the highest sensitivity of 76.4% and the highest specificity of 95% can be achieved.
